First definitions: "that": "used to identify a specific person or thing observed by the speaker" and "is": "third person singular present of be.", wherein "exist" could be substituted for the word be.
Thus,
That that is, is. = the person or thing that is (exists).
That that is not, is not. = the person or thing that does not exist, is not there.

Solving this was intended to be difficult for someone who speaks English as a "first Language".
However...
It may make a lot more sense if you actually read it "Out Loud" with appropriate pauses for the punctuation.
