The first person to punctuate these words correctly wins a BurnAware PRO license:

This  appeared in the 1968 movie Charly, to demonstrate the importance of punctuation.
There are at least 3 possibilities, each with a different meaning:

First definitions: "that":  "used to identify a specific person or thing observed by the speaker" and "is":  "third person singular present of be.", wherein "exist" could be substituted for the word be.

Thus,

That that is, is. = the person or thing that is (exists).
That that is not, is not. = the person or thing that does not exist, is not there.

Well, I tried to understand, but it seems that this is a phrase that sounds ... Greek to me!!!  ;D

Solving this was intended to be difficult for someone who speaks English as a "first Language".
However...

It may make a lot more sense if you actually read it "Out Loud" with appropriate pauses for the punctuation.
